Mechanism of Action:

Gentamicin is an aminoglycoside antibiotic effective against bacterial infections. Aminoglycosides exert their antibacterial action by irreversibly binding to the 30 subunit of the bacterial ribosome and thereby interfering with several mechanisms in the mRNA translation process.

 

Pharmacokinetics:

Absorption from IM injection sites is rapid and nearly complete, except in severely hypotensive animals. Peak blood levels are usually achieved within 30-90 min after IM administration. Because of their polarity at physiologic PH, the Aminoglycosides distribute into the extracellular fluid space with minimal penetration into most tissues except the kidneys and the endolymph of the inner ear. They are eliminated unchanged in the urine by glomerular filtration, with 80-90% of administered drug being recoverable from the urine within 24 hr of IM administration.

 

Clinical Indications:

Gentamicin is used for soft tissue infections, endometritis, metritis, and pyometra. It is also used in the treatment of bacterimiea or septicemia in neonatal foals. Gentamicin has been used in cattle for treating acute colifrom mastitis.

 

Direction for Use:

5 mg per kg bodyweight, beginning day every 12 hours then every 24 hours up to 3-5 days.

 Administer by slow IV or IM injection.

 

Adverse and common side effects:

Gentamicin should be used with caution in animals with compromised kidney function. Avoid concurrent use with other Aminoglycosides because of the potential additive toxic effects. The drug causes transient pain on injection. Ototoxicity can occur but is difficult to detect clinically. Weakness from neuromuscular blockade can occur at high doses.

 

Drug Interactions:

Concurrent use of furosemide can increase the ototoxic potential. Dehydration can predispose to renal tubular damage. Gentamicin is not to be mixed with any other product.

 

Withdrawal time:

Gentamicin is not generally indicated for use in food producing animals.

 

Pharmaceutical precautions: Store below 30oC, protected from light. Do not freeze.
