Pharmacology:
Histamine is are of a number of endogenous substances, autacoids, mediating anaphylactic, hypersensitivity or allergic reactions, other agents are involved in those reactions in different organs to varying degrees according to species.
Release of histamine from mast cells and basophiles occurs without destruction of the cells in physiological immune/ allergic reactions. In severe reactions, or in cases of physical or chemical insult (e.g.bums, wounds, cytotoxins, venoms ) larger quantities of histamine are released by disruption of the cellular, membrane and further release from other body cells ours, particularly in tissues containing smooth muscle. Antihistamines act by competitive antagonism of histamine at the receptor site. They do not inhibit histamine release. The clinical efficacy of the antihistamines aries from their longer half- life, histamine has a half- life of only one to two hours, while the half-life of tripelenamine hydrochloride is approximately six hours following intramuscular injection. Clinical signs of excessive histamine release include increased vascular permeability leading to oedema, bronchoconstriction, hypotension, and stimulation of sensory nerve endings  resulting in pain and/or pruritis.

 INDICATIONS:
Tripelennamine is indicated for conditions requiring antihistamine therapy as follows:

 Horses: Laminitis, feed allergy, insect stings, pulmonary oedema, urticaria

 Cattle: urticaria, asthma, anaphylactic shock, toxemia associated with bacterial infections Or digestive diseases.

Dogs & cats: Eczema, asthma, allergic conjunctivitis

 DOSAGE AND ADMINISTRATION:
Administer by intramuscular injection only.
Do not exceed maximum dose.
A general dose rates = 1 mg/ kg , which is equivalent to :

 Horses & Cattle: 10 ml per 250 kg bodyweight ~ 1 mg per kg body weight, 2-3 times in day.

 Dogs & Cats: 0.2 ml per 5 kg bodyweight ~ 0.5 mg per kg body weight 2 times in day.

 CONTRAINDICATIONS:
Not to be used in animals sensitive to tripelennamine.

 PRECAUTIONS:
Tripelenamine is a CNS stimulant if administered in excess of recommended doses or too rapidly, eg by intravenous injection.

 WITH HOLDING PERIOD:
Meat: 7 days
Milk: 2 days.

 STORAGE: Store below 30°C. Protect from light. Do not freeze.
